I have seen the colony characteristics of one of the Macrophomina isolate collected by me behave different from other isolate.
It is very slow growing in the zig zag pattern. Can I guess, hypovirulent isolates of Macrophomina or it require some test.
What simple test I can perform?
Please those are expert in this field give answer.

I work on chestnut blight fungus Cryphonectria parasitica, hypovirulent isolates. After purification and characterization the isolates, you may apply artificial inoculation the infected tree(case of chestnut), or add to soil with some enhancer for competitions in your research case. the growing form not enough to judge the viral incidence, it may different Macrophomina spp. Or mixed isolate, the word simple test depend on your laboratory facility and budget . Mycoviruses associated with hypovirulent phenotypes have been reported for many plant pathogenic fungi. Common techniques to detect mycoviruses depend on the presence of dsRNA elements. These techniques require cultivation of the host fungus, extraction of nucleic acids and purification of dsRNA.
Researchers, developed a simple and rapid method detect Magnaporthe oryzae chrysovirus 1-A by direct one-step RT-PCR in samples picked using a sterilized toothpick from fungal colonies growing on plate media. This method could be applied for direct detection of mycoviruses from lesions caused by virus-infected plant pathogenic fungi.

you should test it on different plant cvrs at different conditions in laboratory to simulate field conditions (e.g. nutrition, temperature etc.). Preinoculate with your "hipopathogen", than apply different concentrations of pathogen inoculum and see, what will happens. Good luck.
