conventional AMES (MUTAGENECITY ASSAY) assay use vogel bonner salts, spizizens salts etc for prepration of the minimal glucose agsar plates. Is it ok to use M9 salts in this formulation instead of the above two ? will this effect the strain growth ?

composition of the salt solutions are as follows

Vogel Bonner 50X for 1 Liter

Warm dd H2O (45°C) 670 ml,

MgSO4 • 7 H2O 10 g

Citric acid monohydrate 100 g

K2HPO4 500 g

NaHNH4PO4 • 4H2O 175 g

Spizizen's 5X diluted salts solution contains of (per liter):

  • K2HPO4, 14.8 g/l (85 mM)
  • KH2PO4, 5.4 g/l (40 mM)
  • (NH4)2SO4, 2 g/l (15 mM)
  • tri-Sodium citrate.2H2O, 1.9 g/l (6 mM)
  • MgSO4.7H2O, 0.2 g/l (0.8mM)

for 1X M9 (per liter): 0.3% (22mM) Potassium Phosphate (KH2PO4) (3.0g/L) 0.6% (22mM) Sodium Phosphate (Na2HPO4) (6.0g/L) 0.5% (85mM) Sodium chloride (NaCl) (5.0g/L) 0.1% Ammonium Chloride (NH4Cl)(1.0g/L) 0.2mM Magnesium Sulfate (MgSO4) 0.1mM Calcium Chloride (CaCl2) (15.0mg/L) (optional) 0.4% Glucose (or Glycerol) (4.0g/L) (optional)
