I am not quite sure whether I understand your question right. It doesn´t matter whether you separate the proteins in a vertical or horizontal page gel. For the transfer onto the membrane you need another device, either a blotting chamber or a device for semi-dry transfer.

Of course you can. There is no problems running proteins in  horizontal or vertical electrophoresis.
