If using absorbance, would need to blank absorbance system to water or buffer (match closest to diluent in sample), and then could read filtrate. You would only be able to compare sample against blank.

Standard curve is necessary for comparison. If filtrates are routine samples in a process, then it is better to have the standard curve and evaluate the concentrations. Otherwise also, for one single sample, standard curve is a necessity.
