You might be able to estimate it by a noninvasive imaging technique such as MRI. Alternatively, you could use histology (undecalcified methymethacrylate method) and cut serial sections. Unfortunately, the answer to your question is therefore not very easily. We can inject 50ul of fluid into the synovial space of a rat knee which extends the joint capsule, so perhaps 10-15ul would be a reasonable estimate. At necropsy, 200ul of saline injected into the knee recovers about 100-50ul of fluid with an increased viscosity which can be assayed for inflammatory mediators.
