There is not enough dextrose in LB and it's less rich than YEP. But if you supplement LB with 2% dextrose it should be fine for yeast though not optimal.
I don't think it is a good idea. LB has no sugar added and contains 1% NaCl - even if the yeast do grow, the growth will be much slower than on YPD, and they will behave differently (no glucose repression, active osmotic stress response).
LB is specially for bacteria. in case of yeast, it is utilizing sugar in more higher rate as compare to bacteria. and in Luria broth, low sugar concentration and low YE is present. if you are supplementing YE and sugar to LB so its better to use YEPD.
one more thing if you are going for fermentation go with salt media.
Isn't there someone around that can help you with the single components required to prepare YPD? Are you indeed bying ready-prepared YPD? Composition per liter: 10 g Yeast extract, 20 g Peptone, 20 g Glucose. Yust solve and autoclave...
Your yeast strain may be a bit picky concerning vitamin supplements. These are all in the yeast extract, but not not really present in LB. Therefore, it may work when you supplement your LB with Yeast extract, but I would recommend not to use it.
To try for fun is OK, you will get growth but the media is not optimal and therefore you could not compare the results obtained with those from experiments using real YPD.
By the way, I believe all the media we use is good for obtaining high numbers of cells but that doesn't mean they are the closest replicas of what goes on in nature outside the lab. Different growth media leads to different phenotypes. In the case of human fungal pathogens like Candida albicans we adopted the use of YPD and it works well but nowhere in the human body you can find conditions like resembling YPD. Having said that, S. cerevisiae is a different story and probably YPD is a good choice.
I wouldn't expect very much of this medium as for using it for growing yeast. During my researches carried on non-conventional yeast I would always prepare the YPD medium by myself in the way Matthias Brock mentioned. It worked quite well for me, therefore I think it would work fine for you as well. As Matthias said, yeast extract is really of vital importance here.
Hello. As Matthias Brock said, if you have the sole components for the broth, you should prepare it yourself just with Glucose (2%) but no Salt, and I would say it really should give good results with tryptone instead if you don't have peptone. Regards.
