Hello, I want to ask if we can get the data on each pixel point from the confocal microscopy(e.g A1MP Nikon) for an image. Usually, it is in the form of 2400*3400*8units. Can we reshape that data? Can we get intensity from fluorescence or confocal microscopy in the form of a data matrix like we used in Matlab??

let's say for example I have achieved the intensity data from the Matlab in data matrix form, then how can I calculate electric field components Ex, Ey and Ez? Can you suggest me any literature or would like to explain?

intensity data got from Nikon Microscope. This is polarization excitation light intensity? but from that intensity how can I find Ex, Ey and Ez at each point of the data matrix i,e on each pixel.
