ELISA protocol called for me to make a large batch 1X wash buffer from 1% tween in 20x PBS. It says it's stable after 30 days, but its been a week or two over that. Do you think it's still safe to use?
When in doubt always make fresh buffer. The time spent making fresh buffer is way better than assuming the buffer is fine and end up having to repeat the experiment.
Both PBS and Tween are itself very stable reagents that can be stored for long term at room temperarture therefore if you worked clean and you are sure that no contaminations happened during buffer trasfer, you can certainly use it.
if you would be sure about sterility filter it at 0.22um but afterwards it will be ok!
As Manuelle said, PBS and Tween are highly stable, but I would rather keep the at 4 °C. I've had similar solutions stored in the fridge for over 4 months. However, always check for the presence of particles. If you are in doubt I'd recommend you to filter it with a 0.25 um mesh membrane and measuring the pH. If the later has changed, it might mean degrading or presence of microorganisms.
