Currently, I am conducting research on the sensitivity test of gonorrhea bacteria with ceftriaxone combined with betel leaf extract. However, the laboratory where I conducted this research did not have a CO2 incubator, after I did some research and one of them is listed on your website regarding the use of the jar candle method, I have several questions regarding the technical use of this method using an ordinary incubator.

1. What is the application of this method, by placing a petri dish in a jar, then letting it stand for 24-48 hours at 37 degrees Celsius in an ordinary incubator? or just a few hours, then the petri dish is removed from the jar, then incubated without using a jar?

2. In several articles I found information, that the use of the jar candle method also includes moistened tissue then placed under a petri dish in the jar, to provide a supportive environment for the growth of gonorrhea bacteria, is this information correct?

3. When taking samples, I use the Amies transport medium, how long can the gonorrhea bacteria survive in the transport medium? and at what temperature can I optimize the sample storage??
