I am using streptomycin to kill the extracellular bacilli while infecting THP1 derived macrophages with M.tb. Can streptomycin cross the macrophage membrane ?. I read some articles. It says that transformed cells can be selected by gentamicin i.e it kill the non-transformed cells. Both gentamicin and streptomycin are aminoglycoside family. Is my protocol correct?

Krisna Moorthi P Although gentamycin is the most common antibiotic used in invasion assays, the choice of antibiotic depends on the bacterium to be killed. Therefore, for killing M. tuberculosis bacilli, streptomycin could be a good choice.

Streptomycin (in combination with penicillin) is commonly used in cell culture to prevent (myco)bacterial contamination (including M. tuberculosis).
It was demonstrated that streptomycin could inhibit growth of M. tuberculosis within macrophages. Aminoglycosides are incorporated into phagocytes when these cells are in contact with the antibiotic for prolonged periods (i.e., more than 24 h) (Maurin M, Raoult D. Use of aminoglycosides in treatment of infections due to intracellular bacteria. Antimicrob Agents Chemother. 2001 Nov;45(11):2977-86).
As an aminoglcoside, streptomycin targets the bacterial ribosome. However,
aminoglycosides also seem to impact the eukaryotic translation mechanism to promote read-through of premature termination codons (PTCs) in mRNA. Aminoglycosides are therefore considered as potential therapies for PTC-associated human diseases.
