In the articles I have seen, squalene has only been used in vitro, but I want to spray squalene on the leaves of the plant (in vivo). I do not know if I can do this or not.
Look like it is ok to spay on plant leaves. See this patent below. What are you using it for? Kill plant mites?

Abstract
The invention provides a miticide for killing plant mites, which is prepared by taking squalene as a main material and adding a solvent, a solubilizer and/or an emulsifying agent, and is a harmonious pesticide which is safe for human and livestock, is environment-friendly and has deep profound significance for long-term development of ecological environment, the human health and comprehensive treatment of diseases. Compared with the traditional pesticide, the plant miticide of squalene has the following characteristics and advantages that: 1, the squalene is abundant in nature, the content is higher especially in plants and animals, the toxicity thereof is low, the squalene is easily degradable and is safe for the ecological environment; 2, the extracting method of the squalene is simpler, and the squalene can be extracted by techniques of supercritical CO2 extraction and the like; 3, the squalene also can be synthesized artificially, has low cost and is easily used for industrial large-scale production; and 4, due to physical and chemical properties, the squalene can be dissolved in a plurality of solvents, is easily mixed with other mite-killing components for preparing pesticide, and can play a role in improving mite-killing effect."
I do not intend to use squalene for killing plant mites. It is important to me that squalene can be absorbed through the plant leaves and as a precursor affect the synthesis of metabolites.
When you said "Squalene has been only used in vitro", what did the authors do with adding Squalene in vitro? Here is a paper: "Methods for Obtaining and Determination of Squalene from Natural Sources". You can ask those authors. They might be able to answer you some questions.
