In the past we have used Diasorin's ACTH IRMA for assays in human and rhesus plasma. This kit has been discontinued, and I am looking for a replacement. I have tried MP Biomedicals RIA and MD Biosciences ELISA. The ELISA has given more comparable results, but many of our samples are near the limit of detection which results in high variability in duplicate measurements. Does anyone have any other suggestions for a replacement plasma ACTH assay?
i faced this problem in using ELISAfor some cytokines, the problem is that the stander concentrations of the kit has big variance for example the first two high known concentration always you cant be useful for it and you unknown data always plotted in around the forth and fifth known concentration so try do neglect the first two high concentration and make other concentrations for example if you have concentrations like that
2000pg, 1000pg, 500pg, 250pg, 125pg, 62.5pg, and 31.25 pg try to change it to for example to
250pg, 125pg, 62.5, 31.25, 15.625, 7.8123, 3.906 by two fold dilution and I'm sure your problem will be ended

we used recently in our investigations in human pituitary glands the antibody provided by Abcam (Abcam AB8615) - worked fine at least in WB!
