Recommended cell culture conditions:

- Caco-2 cells are being grown in EMEM media + 20% FBS

- HT29 cells are being grown in McCoy's 5A media + 10% FBS + 0.21 g/L L-glutamine

I want to co-culture these two cells together and have found that papers have done this in DMEM media. I would appreciate any advice on how I can get to this stage. I have tried completely switching to DMEM, however the cells did not survive. I have also recently started growing flasks in a mixture of DMEM and EMEM as a way of introducing it slowly, and have found that 25% DMEM and 75% EMEM containing media allows for good cell growth (attached as "healthy") whereas 50% DMEM and 50% EMEM causes the cells to die (attached as "unhealthy").

Finally, I also have been seeing what I think is contamination (attached as "caco-2 cells") but can anyone confirm if it is contamination or dead cells? Would the brown coloration on images 1 &2 also be contamination or something else?

Thank you in advance for your advice/clarification, anything is appreciated!
