Have you tried IS6110 ? is present in multiple copies in most TB strains. RD9 is another good one for MDR strains since is not always in M. bovis.

If you are doing sequencing of these genes, you don't need a positive control, you can align your sequences with availaible genes sequences in ncbi database or check for known mutations confering resistance in this TB drug resistance mutation database:

For pncA mutations confering PZA resistance you can use M. bovis BCG that is naturally resitant to PZA and harbor a mutation at position 169.

If you prefer to use a positive control you can ask some labs for DNA samples extracted from resistant strains or recombinant M smegmatis.

Actually i need positive samples (DNA) of these genes, to see the band while running the samples and these +ve control and comparing each other. I need these positive controls to do PCR and gel electrophoresis.
