I am having some troubles working with a cells experiment and the microscopes. I do not really understand how the Doxorubicin can show flourescent interaction. In some papers, they also show images where cells have blue nuclei (DAPI blue) and red nuclei (containing Doxorubicin).
Also, how about the confocal Images? And, what is the difference between two type of DOX?:

Doxorubicin itself has a intrinsic fluorescence that helps detecting the molecule. As a intercalating anti-cancer agent it has to be in the cell nucleus to be effective. As DAPI also stains the nucleic acids in the cell nuclei. The colocalization of both fluorescences indicates that the doxorubicin is in the cell nuclei where it can show its effect.
But what is your question?
Oh, I also wondered about the intrinsic fluorescence that DOX has or not have. Thanks for your clear answer. OK ^_^ I got the answer. DOX and DAPI, that provides red and blue color in images (confocal and fluorescent images). Question: which one is better of two kinds of DOX (links above): I often use the 2nd one, but I realize the 1st one has "suitable for fluorescence" phrase and cheaper, (need your oppinion) ... Thank Robert !!!
I would purchase the cheaper one, since both DOX have the same purity (98-102%) and although one doesn't say its suited for fluorescence both substances should fluorescence as stated in the description tab on the left side.
To be sure, you could also ask Sigma what they recommend for your experiments!
