Hi Ute, thank you for the protocol. I followed the same protocol. However, the stain gets washed away when I put it in 70% ethanol.

Look into Romeis, old but extremely good for classical colouration.

I must say I chuckled a little reading your problem. My lab has used 1% cresyl violet for years to stain cell nuclei on flat mounts of corneal endothelium. After staining and a water rinse we would process the slides throught 70% ethanol to differentiate the stain. No problems, very nice nuclear staining. Then after many years, we ran out of the stock bottle. Our stain was purchased from Kodak Chemicals. Unfortunately, they didn't sell it any more. We went to another supplier and instantly encountered the same problem. 70% ethanol removed the stain. We bought another batch from another supplier, same problem, A third batch from a third supplier proved no different. I am not certain how the Kodak stain differed from the new stains (they are all certified) but the latter are all removed by a 70% ethanol rinse. I would suggest that after you stain in cresyl violet, rinse in water and mount your slides in a water soluble mounting medium or in glycerol based mount, avoiding the ethanol rinse completely. Alternatively, after the rinse if you can dry your sections without them cracking, you can then mount them in Permount or a related mounting medium.
