I am preparing a Lactobacillus casei cell wall extract (LCWE) to generate a mouse model of Kawasaki disease. I have a query that some researchers go directly to a sonication step after overnight SDS treatment while some researchers go for sequential incubations with 250 mg/ml RNase, DNase I and trypsin after SDS treatment and then proceed for sonication.
I need to know which protocol is better and why? I also need to know where to store the prepared extract?

Dear Rakseh,
Here, we are starting now with the optimization steps for this protocol. Have you already performed a succesfully extraction of the LCWE? We have a lot of problems to determine correctly the protocol of sonication.
