I have applied plant residues (Raw Garlic stalk) with 3 different concentrations in cucumber continuous potted soil. i want to check its efficacy towards microbs addition or not? i want to analysis soil microbs (bacteria, fungi) by high profile gene sequencing method (454 Pyrosequencing method)
1: would this method good to explore the microbial activity under plant residues?
2: if mix the plant residues with AMF fungi, how much this gene sequencing method would helpful?
Dr. Ali, I think you may go for gene sequencing ( 454 pyrosequencing ) method. To answer your second question I should say if there is no host root available within 72 h time, your AM fungi may not survive, therefore, you have to make sure that you have the living plant while applying. Moreover, to get the effect of AM fungi you need to have compatible microbial population. I don't know the major group of microorganisms present in your soil. Accordingly you have to select the AM fungi. For example, Glomus fasciculatum is very compatible with Aspergillus fumigatus. In general, microbial population and diversity is affected by applying different plant residues. It depends upon the quality and quantity of residues you are adding along with soil temperature, moisture ,pH and organic matter content also affect the build up and diversity. In arid soil we are getting on an average 5-30% improvement in microbial population and 7-19% microbial diversity.
