I am looking for a protocol to quantify proteins in the mice brain by western blot technique. I want to see the effect of treatment on AD pathology and neuroinflammation markers. I would like to analyze following proteins: Tau, BACE1, COX-2, GFAP, IL-6, IL-1beta, TNF-alpha, iNOS and p38 MAK. Can somebody share with me the wetern blot protocol with all necessary details. I get a lot of tips from the Abcam company. A better block for me seems to be 2% BSA in TBST as opposed to 5% milk in TBST, and it has always worked better for me to incubate the primary antibody in the blocking solution overnight at 4C. 3x10 minute washes in TBST with mild rocking before and after secondary antibody incubation seems to be good if you use ECL substrates.

I know this thread is a bit old, but I'm curious if Gaurav or anybody else has been successful in detecting iNOS in mouse brain by Western blot.
