I do not sure whether FⅨa is one of the Serine Protease Family. I found the date said FⅩa and FXIa are belong to Serine Protease.
My question is expect the spectrozyme fⅨa that cansed as a chromogenic substrate, S-2288 cansed as a chromogenic substrate?
Factor IXa is a serine protease that cleaves and Arg-Ile bond in factor X.
S-2288 (Sigma-Aldrich) is D-Ile-Pro-Arg-p-nitroanilide.
I don't know how well Factor IXa uses that as a substrate, but it has been reported to use similar synthetic substrates such as CH3SO2-D-Leu-Gly-Arg-4-nitroanilide (see BRENDA database link below), so it might work.

You might also be able to use Z-Lys-SBzl (Sigma C-3647)as a substrate in combination with Ellman's reagent to detect the product.
