From my humbul reading for many articles about Rhumatoid arthritis it mentioned that sero positive patients will never be negative again even with methotrixat treatment but one of our patients showed negative RF and normal Anti-CCP titer after 3months of topical treatment with some herbs.

Rheumatoid may convert from seropositive to negative. It has been reported in a study 2013 published in J Rheumatol that about 18% of cases may convert from positive to negative.

Glad to hear that the anti-CCP is negative as this carries a promising prognosis.
As for your second Q, if you are sure the cases is RA, herbs alone do not have a primary role (even if orally provided and not just topically).

In your openion If the same treatment applied to another patient and gave the same result, how many patient I need to proove that treatment?
Dear Dr Zina Al-Shami ，I found your case very interesting.And one of my patients also turned to be sero negative with negative RF and low titer of Anti-CCP after 4-5 years treatment. However, if 3 months therapy with herbs could made it happen, I think it is necessary to confirm the diagnosis.
