In order to confirm viability of cells, I used vital dyes like FDA, resazurin. I got positive for that at the same time cells have shown positive for PI as well. If my cells are viable, can they take PI also by any chance. Or can PI pass through the membrane via transporters or cells with leaky membranes.

Yes use Alexa IV apoptosis kit. It will mark dead cell and remaining will be live cells. Facs. PI tags or bind with phosphotydal serine. Live cell don’t allow but it can pas dead cells.
