I want to separate T cells from PBMC using a T-rosette. Growth factor is necessary for the cells to present proteins, particularly CD2. Can I use Bovine serum albumin instead of Fetal bovine serum.I am looking forward to receiving your comments.
BSA cannot fully replace FBS if growth factors are required.
BSA is a major component of FBS and provides essential functions like binding and transporting molecules. But if you use BSA instead of FBS, it won’t provide the broad range of growth factors, hormones, and other nutrients which FBS can provide.
So, BSA is not a suitable substitute for FBS, if you are interested in growth factors.
Based on my experience, using Bovine Serum Albumin (BSA) instead of Fetal Bovine Serum (FBS) for T-rosetting is not advised. FBS contains many growth factors vital for maintaining cell surface proteins like CD2, which is crucial for the rosetting process. Substituting with BSA, a single protein, usually leads to poor results. For best T-rosetting, stick with FBS. If you're concerned about FBS, consider serum-free media with specific growth factors. While short incubation might allow for slightly less FBS, replacing it entirely with BSA risks CD2 presentation. Overall, BSA cannot effectively replace FBS for supporting cell health and CD2 presentation needed for successful T-rosetting.
