Generally, we maintain 16h photo period in culture room for plant tissue culture. But continuous 24 hrs light is also beneficial for plant growth. So, for an experiment can I use 24 hr photo period? I need suggestions
Anjum Ona I would not recommend it. It also depends on what plant species you have- a short day plant or a long day plant. Some might hinder plant development. Here is a review paper for you.

Anjum Ona I also agree with Yuan-Yeu Yau . continuous exposure of light onto a in vitro plant may cause a reduction of photosynthetic efficiency and also plant growth. If you want to understand the light stress mechanism of tissue cultured plant/ callus you can apply the 24hrs light and unravel the new avenues.
In general 16 hrs photo and 8 hrs dark period is better for plant growth. further Over light exposure inhibits/reduces the cell division and differentiation in in vitro culture-derived plants.
Dear Anjum, most of plants follows circadian rhythm for their development. If it is Long Day or Short day plants you need to maintain light condition as per the plants requirement and your experimental objectives. In case of day neutral plants viz Banana you may go up to 20-22 hr light and 2-4 hr dark's for fast development. Prolonged light is not at all desirable for any plant its induces the expression of genes involved in the stress

I also agree with Vijayendra Dalvi that most of plants follows circadian rhythm for their development. Here we need understand that the nature of the plant (its photoperiodic requirement matters). Since we're not interested to get flowers from in vitro plants the effect of 22-24 h light may not be our concern. However, the dark period is also vital for respiration of plants. In some (C3) plants photorespiration can take place during the light hours.
