I want to transform bacillus subtilus with ligation reaction directly. Is that possible?
To be more clear, I have a shuttling plasmid between bacillus and e.coli. I know usually we need to do the construction in E.coli, extract plasmid and then transform to Bacillus. But my plasmid seems like have toxicity to E.coli. So I want to skip the E.coli and do cloning construction on Bacillus directly. Would that be possible?
