We have prepared the paclitaxel loaded Liposomes. Paclitaxel is water insoluble. We want to separate the untrapped paclitaxel from the liposomes. for the same can i use 0.8 µm syringe membrane filter? in the article enclosed they used it , but I am little skeptical about. Also looking for the suggestion about the use of centrifuge to separate the untrapped paclitaxel.
Filtration is better than the centrifugation on all practical aspects in your case. What hurdles you see in the filtration, authors have used it well!
There are two things that we are talking about. One is the particle size and one is the molecular size. The molecular size or wt of PTX is close to 850 Da whereas the molecular size of lipsomes will be several thousand Da. If the particle size of Liposome is less than the pore size (800nm) then you will loose your sample. Some lipisome/lipid will also adhere to the membrane filter which will lead to loss of drug loaded liposome. You may try a 100nm or 200 nm pore membrane filter or if possible a better way would be to try a dialysis membrane/dialysis bag of 5 or 10 KDa. PTX should be soluble in large dilution in water.
