Can I increase The extension tempertaure of my Taq polymerase (Quick-Load® Taq 2X Master Mix from NEB) ?

I did a temperature gradient PCR where I got specific band at 68 degrees.The extension temperature is also 68 degrees. When I run the product on gel I got a less intense amplicon for 68 degrees compared to other products.Is this because of the same annealing and extension temperature.Can I increase the extension temperature because the Taq polymearse I am using is recommended by the company (NEB) to be used at 68 degrees extension?Kindly help me in this regard.

Hello Risha,

I think you dont need to increase temperaature, how about cycles ? how namy cycle did you run ? if posisble increase your cycles
