Is there a standardised protocol available for this method? Has anyone tried it with samples containing nucleotides or proteins? I already have reference paper but insufficient information. Ref 1 - Burton K. (1959) Biochem J.

Why don't you try a Mg ion-selective electrode to estimate free Mg in protein containing samples. This is because any chemical precipitation or titration technique will disturb the equilibrium existing in solution between complexed Mg and free Mg and as such your estimation will give rise to total Mg only.
