Hi, I want to do an in-house TSA for IHC using (primary Ab then) a standard biotin conjugated secondary antibody, then avidin-peroxidase, then tyramide-biotin, then avidin peroxidase, then DAB. Will this work?

I'm basing it on this protocol https://sites.google.com/site/helobdellaprotocols/histology/tsa

Also wondering if the overnight wash after tyramide-biotin can be avoided? There seem to be stop reagents in some kits, what are they and how might one make them in-house?

Thanks, Alan
