Is it safe to analyze the ‘temporal change in plant species composition’ using the data sets generated from two different sampling techniques, i.e. Phytosociological relevés (0-7 cover-abundance scale) and sample plots of 100 m2 (0-7 cover-abundance scale)? OR does it need any standardization before making comparisons?
Here, the same landscape was surveyed at different times using different sampling techniques, but was not plot-to-plot re-sampling!

I believed sampling bias would led misleading inferences. Balanced and standard samplings will result meaningful reasoning.
I guess you can. but better to compare the sites using shanon and Jacard indexes.
try to use Sigmastat software or Distance.

You can use a rarefaction analysis to compare individual-based interpolation and extrapolation of species diversity for Hill numbers in different sized samples. R program (iNext package) is very useful.
