I am working on the microbiological characterization of kombucha and I need to extract DNA for a metataxonomic analysis. In my lab, we have the Wizard® (Promega) commercial kit for DNA extraction, and I would like to know if anyone has experience using it with kombucha.
Is it possible to extract DNA directly from the liquid sample by centrifuging it to obtain the cell pellet? Or would any pre-treatment (e.g., filtration or enrichment) be recommended?
I would appreciate any advice on optimized protocols or specific adjustments for this type of matrix.
