I have three kelp, two green, and one red seaweed species that I would like to analyse and quantify for their fatty acid content. I do not intend to differentiate between lipid classes. Would a direct methylation approach be suitable for this purpose? I plan to use freeze-dried, powdered seaweed and extract the lipids using 0.5 M NaOH in methanol, with C21:0 in chloroform as an internal standard. For methylation, I will use 5% sulfuric acid in methanol and heat the mixture at 70 °C for two hours. The resulting FAMEs will then be extracted with hexane and washed with saturated sodium chloride solution to remove residual aqueous components and impurities.
