Hello..You can just try only urethane, but take care about the dose as well as time, do your experiment as soon as possible after giving urethane.

Hi, yes you can definitely try to use urethane alone, but also bear in mind that urethane is thought to be cancerogenic and can only be used in acute experiments. I know that several groups used urethane alone i.p.  in mice for measurements of neuronal activity where it seems to give stable readouts. But also recently it was used for fMRI studies. (e.g. Schroeter et al., Neuroimage 2014 or Grandjean et al., Neuroimage 2014). But like with all anesthetic protocols, best is to test it first with your given setup, readouts and also animal strains and only then you will clearly see if it works for your purposes.

urethan and alpha-chloralose are used together to lower the doses injected. Urethan used alone required high concentrations which interfers with nicotinic, acetylcholine and GABA receptors and inhibate NMDA receptors (Koji Hara, 2002 ). It also causes respiratory depression and mucus build up in the lungs so even if you intubate the gaz exchange will be affected and the PCO2 will be very high which will change your CBF results. Alpha-chloralose has a low cardiovascular impact but is not stong enouh to be used alone.

I would do more literature search before using alpa cloralose with or w/o combination with urethane.
