I am working on Phytophthora and am currently trying to maintain a population of 5000, 15000 and 20000 Sporangia per mL. I tried harvesting Sporangia using Petri solutions and could only maintain 5000 and 10000 Sporangia per mL? Any suggestions on how to maintain 10000 Sporangia per mL?
You can try infecting susceptible A. thaliana strains and then harvest them once fully infected. Check for sporangia formation under 4X or 6X, vortex them in chilled water and strain with cheese cloth. Should get 10^6-8 count/ml.
If you can grow it on solid medium into a Roux bottle you can try to harvest with a saline solution and glass beads
We regularly grow P. infestans on Rye Sucrose Agar (RSA) and harvest zoospores from 12-14 day old plates by flooding them with ice cold water and incubating 2-3 h @ 4'C. You should get plenty of mobile zoospores. If you start a new RSA plate by plating zoospore solution you get a very synchronised sporulation which will increase your yield even further.
