Hi,

I have recently thawed a vial of induced pluripotent stem cells on geltrex matrix membrane. I have used ready-to-use geltrex matrix solution to coat the well of a 6 well plate. Once a layer of gel is formed, I removed the geltrex solution and added 1 ml of essential-8 medium and thawed a vial of iPSCs exactly as per manufacturers protocol into the pre-coated well . All the reagents were from Thermo fisher scientific.

After 24 Hours, I could barely see any cells stuck to the basement membrane and performed a partial media change everyday for 3 days and there is no sign of a single iPSC cell colony. This is the first time I'am initiating iPSC culture and any suggestions would be much appreciated.
