I have tried administering rotenone intraperitoneally to C57Bl/6 mice in my lab, but there is high mortality rate. Subcutaneous rotenone at a dose of 2.5 mg/kg for 28 days did not show any changes in alpha synuclein levels. I m in need of an acute rotenone model of PD, in which I can check the effect of molecule against alpha synuclein aggregation.
Monika Sharma, at this point, I think it would be more accurate to describe your experiment as an acute rotenone toxicity assay - not a 'mouse model' of Parkinson's disease. It may be worth considering the following possibilities:
I say this because there is a great tendency in biomedical research to uncritically build up a weak and uninformative 'disease model' paradigm. This creates an illusion of certainty and causality that is really a house of cards. Such research provides no benefit for real people suffering from disease. It would do far more good if you were to simply publish your atypical findings with rotenone rather than to seek out a confirmatory scenario.

I have the same problem. I have tried 28 days i.p. in C57Bl/6 mice dose 1, 2, 3 mg/kg dissolve in DMSO and sunflower oil as previous studies have done. It seem not difference, but i have choose 3 mg/kg. I think I might extend the rotenone treatment to 42-45 days as your suggestion. Arman Rahimmi
