I want to express shRNA against my gene of interest in a tetracycline regulatable manner (tet-on system). Can anyone suggest some suitable plasmids for the same? I have shRNAs with 5'BglII and 3'HindIII overhangs. Are there any relevant plasmids with such cloning sites? If I add overhangs for any other restriction enzymes will it alter the processivity of the shRNAs?

The plko.1 lentiviral plasmid (https://www.addgene.org/21915/) might be of use. however i don't think they have the same cloning sites that you want (They have AgeI/EcorI). If you have an shRNA that you know gives a good knockdown, it can be modified to be ligated into the plko.1 vector
