Can anyone suggest to me a good medium in order to induce sporulation in the fusarium oxysporum culture? I want to harvest spores from this culture in order to carry out an assay.
Most fungi sporulate well on oatmeal agar and cornmeal agar.

There are mediums for fusarium oxysporum that produce good sporulation. You can find them in literature. WA with soil is good medium to produce very good sporulation. if like i could send to you list of mediums. Best regards
Most fusaria sporulate well on tap water agar with a few sterile rice grains placed on the agar. I usually use 4 to a plate. You can place the plate on a microscope stage to check for conidial production. Inoculate the culture in the center of the plate and let the fungus grow from there.
potato dextrose agar but the traditional preparation with potatoes water and agar, the fusarium sporulate better than in the comercial potato dextrose(PDA)

I find that Fusaria and many other filamentous fungi sporulate more quickly if the medium is more dilute than the manufacturer's suggestion, half-strength or even less. This is particularly true for endophytic fungi, which can be osmotically shocked by transfer from the plant to a full-strength medium.
Carnation leaf agar is the best medium for spore productionbut i saw Mungbean agar also as a medium for spore production but we try to do it unfortunatly this last medium did not response
its simple as like and prepare a water agar with pieces of sterilized filter papers placed on the top, inoculate Fusarium on the filter paper, you get sporulation .. try this
The cheapest effective method is to prepare a medium with distilled water plus wheat stem fragments in flasks and inoculate the medium and incubate it under artificial lamp at room temperature. A method similar to that suggested by dear Gunasingham but with wheat stem fragments and water in a flask.
F. oxysporum is an easy fungus to sporulate, use corn meal, oat meal , PD medium but do not add sugar . you might have black light which is far UV light , If so incubate your culture under such light for a week at a 20 degree celsius, you will get a massive spores ,If not the regular incubator is good enough
I have used PDA, corn meal,oatmeal, Sabouraud ,however some species of Fusarium have had good sporulations, but other didnt have same behavior, I recommended you try with changes light and temperature, for example exposure them to light 12 or 16hrs of light, at 5 to 25 degree celsius.
The easiest way is to just use PDA and then scrape the surface for spores. That should be sufficient for most F. oxysporum isolates. In case you are working with a low sporulaton isolate, try cultivating it in liquid Fusarium Complete Medium (CM) at room temperature, low agitation and dark conditions for 5-7 days. You can then collect the spores by filtering through a double layer of cheesecloth and centrifuging to collect the spores.
Fungi in general, and Fusaria in particular, the environmental conditions necessary for good sporulation are more limiting than are those growth. A number of microbiological agar media such as Malt Extract Agar, Oat Meal Agar, etc. are useful for Fusarium sporulation. However, Potato-dextrose Agar is more efficient in the sporulation of Fusarium. Other natural media such as cereal straws, seeds embedded in water agar, green carnation leaves, and Nash medium are excellent for asexual sporulation in Fusarium.
I think use PDA with additional supplements. like 1 gm of glucose, peptone, yeast extract. Add nitrogen supplement in the media too. Like ammonium tartrate (0.5 M 5-to 10 ml /L of media) and maintain the pH of the media upto 6.5. I hope that will enhance the sporulation. Best wishes
Maximum mycelial growth was obtained in potato dextrose agar as semi-solid while Maximum sporulation will obtained in oatmeal agar and mycological broth. Please see the attached references
What about the wheat blast fungi sporulation? How can the rapid sporulation of wheat blast fungi be induced? Is there any other way other than using different culture medium?
