Extracellular vesicles (EV) is the current term of vesicles isolated from bodily fluids or culture supernatant. EVs are comprised of vesicles ranging in diameter from 30nm -1µm. Exoquick is developed to isolate vesicles from 60-150nm, however this is measured using NanoSight methods which is not optimal for the measurement of larger vesicles. I am convinced larger particles are being precipitated too by ExoQuick, however i have hard time getting evidence. Anyone able to provide me with literature on the size distribution of ExoQuick precipitated human serum vesicles? or a FACS analysis picture?
If you are interested in isolation of exact sub-population of exosomes from other extracellular vesicles using ExoQuick, I recommend you to combine it with serial filtration. If you do serial filtration(e.g. 0.4 um and 0.2 um) prior to ExoQuick, you will be sure that the input material for polymer precipitation is in the range of exosomes and then if you do isolation with ExoQuick the size of particles would less than 200 nm, mostly between 50-150 nm.
