Can anyone please let me know how to prepare metaphase spreads from cultured CD4 and CD8 lymphocytes invitro?

We isolate and sort the cd4 and cd8 population from whole blood and enrich these population by further culturing these cells in vitro by adding mitogen PHA followed by colemid addition (10ug/ml) after 69 hrs and harvest at 72 hrs for metaphase preparation. It would be highly appreciable if anyone help me out in this regard.
