Can anyone please help me in estimation of amylose using 1N acetic acid and iodine solution ?

In estimation of amylose (Juliano, 1971), I used 1N acetic acid but got color developed only in a few samples. And then I used glacial acetic acid (99.5%) in another set of samples and got the color developed in all samples.

How does this affect my experiment ?
