Has anyone tried fixing cells in an MSD plate? We normally use 80% acetone but I don't know if this will interfere with MSD detection. I would also like to use the same antigen in a sandwich assay. I was planning to use Triton-X to permeabilize the cells. Has anyone used Triton before with MSD?

I don't know MSD, but acetone react more or less with the TPP's plate we used.
Try 4% formaldehyde or paraformaldehyde solution in PBS. It should work well enough. Triton X looks OK, but you can use Tween-20 as well.

Never tried fixing cells on an MSD plate, but the acetone will likely destroy the plate material (plastic?). 4% paraformaldehyde (fresh in PBS) (10-15 min room temp) followed by the 0.25% Triton X (5 min room temp) usually work fine. Good luck!
