I'm doing my research on lutein extraction from marigolds, and further its encapsulation.
I have many articles regarding different methods, especially soxhlet, enzymatic extraction and methanol-based extraction. However, I can't get a practical point of doing it, especially after extraction I have to find the concentration of lutein obtained. Paper and HPLC will be used.
But for higher quantities, paper or TLC will be used.
Please help me in understanding the practicality of the work.

Oh dear! it depends on ones own approach, soxhlet extraction method saves time and assure the maximum recovery of the target, if is it not heat sensitive. For lutein use petroleum ether and acetone in 1:1 ratio or you may try dichloromethane as extracting solvent in soxhlet method, and no issue if you latter analyze through HPLC, TLC
The brief steps are:
First of all you isolate the pure fraction from plant extracts using following techniques. First run TLC to know the type of fractions. After that Column Chromatography with HPLC, after isolated pure fraction run FT-IR, LC-MS, HNMR, CNMR for structure elucidation. You can read my papers too.
in soxhlet you can use different solvent for polar and nonpolar
for Polar: water, methanol, ethanol
for Non-polar: hexane
Soxhlet is a simple method of lutein extraction. FT-IR is used to eucidate the structure of lutein.
