I synthesized some hydrophilic polymers with multiple carboxylic acids as end groups. They were cross-linked by coupling with water soluble metal ion in water. These polymer complex formed different vesicle-like structures as detected by tradition TEM (not cryo-TEM). However, DLS can not detect them even at high concentration. The only peak showed on DLS is around 5 nm which belonged to free polymer. My questions are 1. Can DLS fail to detect these vesicles? 2. Can these vesicle-like structures just form after they are dried and so they are just some ring-like stains?
Has anyone had such experience with TEM? (By the way, these experiments were made three years ago and I didn't follow up because of these questions)
