I've been having problems with Arabidopsis growing, since 4 months ago approximately. This is my procedure: I start with sterilization (bleach 50%) and 3 days stratification (4°C in darkness). Then I sow the seeds (Columbia) on MS medium, containing 1% sugar, 0.7% agar and pH 5.7. After 2 weeks (when the first leaves appear), seedlings are transferred to soil (peat moss and perlite), in 2:1 proportion. Seedlings are kept in a growth chamber, with 22-25°C, 40-80% humidity, 16:8 photoperiod and 40 PAR. Everything is normal until here, but around one or two weeks after the trespassing, seedlings started flowering. I thought it was a fungus problem, because the first essay was severely attacked by fungus, but in the second essay (with differents seeds), I took care of avoid the fungus contamination, adding fungicide once per week, but I got the same result. Then, in a third essay, I changed the watering frequency, I started to fertilize (once per week), I changed the temperature (now the range is 20-25°C), and finally, in the last essay, I changed the photoperiod to 12:12, but nothing! seedlings have the same behavior! I know that early flowering can occur by stress conditions, but I don't know what other factor is affecting the growth...Can anyone tell me some idea to get a normal growth? Apologize for my english, I'm learning, or at least, I'm trying.
