My lab is studying DNA damage in breast cancer cells using alkaline comet assays. About half of the comets I would count as comets are not being recognized by OpenComet. Sometimes it won't recognize my comets at all, other times it will only recognize the head. The software is also recognizing random, small points as comets. I have attached some images that OpenComet analyzed to illustrate each problem. I have reinstalled the software to confirm I have version 1.3.1.
Any and all advice is appreciated!
I emailed my question to the developers and they told me that the issues are likely caused by the complex background in my images, the banding patterns. They advised me to look into ways to correct the banding before analyzing the images in OpenComet or find a way to alter the experiment to lessen the bands in the background. I will look into ways to do both, if I find any solutions, I will post them on here!

I assumed it is astronomical Images in FITS format (i.e. images.fits).
In that case you may be stock the same frame images and see. comet will see and rest of the stars moving .
And it is possiable in IRAF software.
by using noao>irred>ccdred>combine
