First the dry soil is mixed with CaCO3 and kept 24 hours at room temperature then the sample has been pretreated by dry heat (at 80˚C for 20 minutes), moist heat (at 60˚C for 20 minutes at water bath), sonication and 1.5%( phenol+ saline solution). Then 100 µl aliquot from 10-4, 10-5, 10-6, 10-7, 10-8, 10-9 dilutions were plated on starch casein agar plate containing fluconazole at a concentration 60µg/ml. But there were no single colony rather a cloudy and turbid growth on the entire plate. I have seen and followed many paper but didn’t find any solution. Please give me some suggestions from your experience. Thanks.
