This is not an easy question. Two methods that can be tried are DIONEX chromatography of an acid hydrolyzate (2M TFA 121C 2 hours) using standards for reference or methylation analysis (permethylation of polysaccharides, methanolysis/hydrolysis, reduction with NaBH4 or NaBD4, acetylation and analysis by GC-FID and GC-MS).

If you simply want to measure total carbohydrate content, the phenol-sulfuric acid method is one of the standard techniques.  Reducing-value based tests like Fehling's test, etc., will not work, since the reducing value of polymeric carbohydrates is very small. Likewise, the iodine test is typically used only for starch determination.

The chromatographic techniques mentioned above are useful for identifying and quantifying the monomeric constituent sugars in your polysaccharide, but are not typically used for simple quantitative measurement of how much material you have.

You may also want to consider methods that measure uronic acid content, if your polysaccharide is acidic (i.e., contains uronic acid groups). The carbazole method is one, others also exist.
