Those are some beautiful histo-slides there. Have you determined the oestrous cycle phase (proestrus, estrus, metestrus and diestrus) in the rats before sample collection? This may greatly assist in order to rule out changes associated with cycling as the rat has a very short cycle (4-5 days). Without determining the exact oestrous cycle period, it may be difficult to conclusively associate the findings to the exposure.

This guide below gives a detailed technique of evaluation the oestrous cycle in rats.

Did you perform hormonal evaluation (estrogen, progesterone, P:E ratio)? This will also give you an insight on the changes and association between the tissue responses seen and the hormonal production profiles. 

The pictures can be interpreted as they are, but there will be no conclusive detail as to whether the changes are influenced by the cycle or not. For example, your exposure may have the potential to alter the oestrous cycle and make it irregular, but without knowing the cycling phase, we may not be able to make that deduction.

Thank you so much for your help, before i was started with experiment i did estrus synachrization and starting exposure at diestrus cycle, FSH , LH hormonal assay were done and i found FSH level was decreased but LH  level still within normal range. 2 rats only at day of collection samples were in estrus phase at period 15 days while at period 30 and 60 days less than 5  rats were in estrus in both periods.

In photographs follicles with different advance in maturity are observed, and there are also changes in the stroma of the ovary.

Germ both stromal cells and are susceptible to hormonal stimulation (FHS). He was waiting for a change of radiation (inhibition or hormonal stimulation) at maturity of the first or second amendment?

I agree with the previous responses. I do have a question though (I might have missed it) - but  do you have ovary samples from  non-exposed age-matched and estrus cycle -matched  (same stage of esters cycle) rats ? Proper negative control samples are required for these slides to be compared the treated ovaries.

Thanks for your interset, yes i have ovarian section from the control group which are non exposed rats. About the estrous cycle, before starting estrous synochrization of all rats in all experiment groups were done and 85% of rats were in diestrous phase when i started the experiment ( i.e. exposure). Regarding to FSH and LH levels also checked by ELISA kits and the FSH level in exposure groups was lower than control rats, inaddetion to that oxidative stress biomarkers MDA was elavated and GSH-PX was lowered and Melatonin level was lowered too compared with control group.  

during taking the samples only three rats from exposure groups were in estrous phase and others mostely were in metestrous and diestrous 

What about the estrus phase at the time of ovary collection (necropsy) ?- this is more important because this is what is on your tissue section.

 I would suggest that you have your slides read by a veterinary pathologist who is an expert in ovarian histology. 

You would also need to take photos of the entire slide of the ovary as well as having several sections (at predetermined intervals) in order to have the entire "picture."

what i am looking for is the pathological changes in ovarian follicles as well as in stroma, ( i.e. is there any deganartion or apoptosis processes in ovarian follicle cells and in oocyt ?) for example this photo showed two neucloulus in oocyte neucluos. Is it normal and i have four sections from exposure groups has the samething.   

As the ovary is an organ that is constantly cycling - its histology will change with the cycles. There will always be apoptosis and degeneration in various stages depending on the cycle as well as the age of the animal not counting the effect of your treatment. You will need to section the entire ovary and quantify the changes (and compare this to the controls) to evaluate the changes correctly. To do this - the pathologist has to see the entire ovary and all its sections - not portions of sections of the ovary.

i already i had whole entrire ovary photos, you mean to compare with low power between photos and then to use high power to see the changes

You have to examine the entire ovary (over several sections) for changes both at LPO & HPO - in addition to identifying the stage of the estrus cycle, make a quantitative evaluation - how many oocytes/follicles (at each stage), CLs, how many apoptotic oocytes, changes in the stroma, identify cysts, identify proliferative changes, etc.  It would be best to ask a pathologist to help you out.

You need to examine both the ovarian stroma and the different stages of ova formation. The best way to do so is to start with investigating the histological sections of control group (at least six rats). You should count the follicles and it is also better to do some morphometric analysis. This will give a good picture for the normal histological structure. Then you switch to the experimental groups keeping in mined the control one. The deviation from the normal architecture can be described as histopathological changes..

Add to full film says Dr. Prague. Photographs shown are ovarian and are stained with HE, the changes listed in them should be compared each group of rats, according to the treatment received. The findings should be related to the maturity of primordial follicles and corpora lutea found. Clear compared with the control group. Hormonal response relationship would be observed FSH / LH
