I have cells that have been infected with mycoplasmas. I am currently treating them with plasmocin.Can my liquid nitrogen tank be infected with mycoplasmas?If yes, how can I clean it?I'm grateful if anyone can guide me
Always leave the tank in upright position. Any tilting, especially with liquid in the tank may cause breaking of the neck and the will be useless. Let all liquid nitrogen evaporate. Remove debris left on the bottom of the tank ( cryovials, tweezers, etc.) ; Rinse with water. Aspirate with a vacuum pump and a tube set on the bottom of the tank. Add diluted bleach and let sit at least one hour, but not more than 2 hours. Swirl gently every now and then. Aspire all the bleach. Rinse with at least 1/2 tank of water once. Rinse with 1/4 tank of water twice . Let dry completely, otherwise you will have a permanent ice coat on the bottom to accelerate evaporation. once totally dry, add liquid nitrogen to 1/3 of tank.
